We report on a synthetic membrane channel made entirely from DNA and anchored to a lipid bilayer via cholesterol side chains, which we created using scaffolded DNA origami. We characterized its transport properties and demonstrated its use as single molecule sensor. Moreover, we report on a solid state nanopore force spectroscopy platform we used to study DNA-histone interactions and the impact of CpG methylation on nucleosome-modulated gene accessibility. At last, we describe a solid state nanodevice to obtain the electrophoretic mobility and recoiling dynamics of long DNA in a single molecule experiment.
