In this thesis, a new method for the isolation of mitochondria from cell suspensions and small amounts of fresh and frozen stored tissue has been developed. Obtained mitochondria exhibited an exceptional quality, regarding their biochemical function, structural integrity and purity. Furthermore, a new approach for the quantification of isolated mitochondria by flow cytometry has been developed, allowing for mitochondrial comparisons based on an equal number of mitochondria instead of equal amounts of protein. This is especially important if mitochondria from different origin are compared as they may differ significantly in their protein content.
