Abstract:
Endoplasmic reticulum- and mitochondrial unfolded protein responses (UPR) in intestinal epithelial cells (IEC) have been implicated in the pathogenesis of inflammatory bowel diseases. This study revealed that an IEC-specific deletion of the mitochondrial chaperone Hsp60 disrupts epithelial homeostasis in both the pre- and postnatal gut. More precisely, HSP60 deficient IEC show an activated mitochondrial UPR accompanied by an impaired proliferative response and a loss of stem cells.

„Unfolded protein responses (UPR)” des Endoplasmatischen Retikulums und der Mitochondrien in Darmepithelzellen (DEZ) sind Teil der Pathogenese chronisch entzündlicher Darmerkrankungen. In dieser Studie konnte gezeigt werden, dass eine IEZ-speifische Deletion des mitochondrialen Chaperons Hsp60 zu einer Störung der Epithel-Homeostase sowohl im embryonalen als auch im adulten Darm führt. Im Speziellen weisen HSP60 defiziente DEZ eine verminderte Proliferation und einen Verlust an Stammzellen auf.
