In this work have been improved the method of the genetic modification of
Gluconobacter oxydans and a selected enzyme of the metabolism were analyzed. For G. oxydans 621H and G. oxydans DSM7145 was established an upp-method, which was based on a 5-fluorouracil selection. By this method were constructed following mutants: transhydrogenase-, putative polyol dehydrogenase-, 6-phosphogluconate dehydratase- and KDPG-aldolase mutants. It has been shown that the Entner-Doudoroff pathway is important for the metabolism of gluconate by the reason that the 6-phosphogluconate dehydratase- as well as the KDPG aldolase mutants could not grow on gluconate-medium.
