Pancreatic ductal adenocarcinom (PDAC) is a highly mortal disease. We developed the tamoxifen-controllable LSL-RosaKE mouse model which enables pancreas-specific expression of KrasG12D from the murine Rosa26 locus to analyze the impact of KrasG12D on tumor maintenance. To monitor tumor cell proliferation in PDAC
in vivo, the LSL-PCNAfLuc mouse model was used. Herein, firefly luciferase is expressed under control of the proliferating cell nuclear antigen (PCNA) promoter. Isolated PDAC cell lines were then used to develop a versatile high-throughput screening tool to identfy and validate potential drug candidates.
