The rise of antibiotic resistant bacteria, such as multidrug-resistant
Staphylococcus aureus, calls for novel treatment solutions. Targeting of bacterial virulence represents one potential strategy, with the ClpXP protease complex of
S. aureus as a crucial regulator. This protease complex was further investigated by chemical and genetic modification. Novel inhibitors of ClpX and ClpXP were discovered and characterized in vitro, as well as in vivo. Genetic modification of
clpP revealed new insights into ClpP mediated virulence regulation.
