The degeneration of midbrain dopaminergic (mDA) neurons is causing Parkinson’s disease. To design future therapies basic research was done. Here, the development of mDA neurons was analysed by using animal model organism (mus musculus). The Wnt signal (canonical wnt signalling) is transduced through frizzled (Fzd) receptors. Wnt1 is necessary and sufficient to develop mDA neurons. Due to expression studies two of these receptors were chosen for analyses. Using biomolecular methods the Fzd3 ko, Fzd6 ko and the Fzd3/6 ko mouse were investigated. Here it was shown that wide redundancies between these proteins were detected. Furthermore these receptors may play a role in PCP signalling. Again, in the canonical Wnt pathway, Lef1 is necessary for the proper development of mDA neurons. This was shown by analysis of the Lef1 ko mouse, were a significant reduction of mDA neurons was seen.
