Abstract:
Dysbiosis and metabolic alterations of the gut microbiome have been described in inflammatory bowel diseases (IBD). The aim of this work is to identify functional signatures associated with disease outcome or response to therapy in two longitudinal cohorts of patients with IBD, and to mechanistically characterize their pathogenic potential using a gnotobiotic humanized mouse model and an integrative multi-omics approach.

Dysbiose und metabolische Veränderungen des Darmmikrobioms sind bei der Entstehung und dem Verlauf von entzündlichen Darmerkrankungen (IBD) beteiligt. Ziel dieser Arbeit ist es durch zwei longitudinalen IBD Kohorten eine prognostische sowieso diagnostische funktionelle Signaturen im Zusammenhang mit IBD zu identifizieren und das Ansprechen auf eine Therapie abzuschätzen.
Um das potentielle pathogene Potential dieser Signatur abzuschätzen wurde ein humanisiertes Mausmodell etabliert. Zusätzlich konnte mit Hilfe eine multi-omics Ansatzes die Funktionalität anhand von zwei IBD Kohorten charakterisiert werden.
