Inflammatory bowel diseases are chronic conditions of the gastrointestinal tract. IKKα kinase is involved in many inflammatory-mediated human diseases. Here, the role of IKKα in intestinal homeostasis is investigated and reported that IkkαAA/AA mice are highly susceptible to DSS-induced colitis. The phenotype is rescued by loss of caspase-12 or exogenous administration of IL-18. Importantly, defective IKKα activation impairs autophagic protein degradation and elevates ER stress. Thus, IKKα is identified as a novel link between autophagy, ER stress and caspase-12 function.
