The role of IKK2, TAK1, TPL2 and TNIK, individual components of the LMP1-CTAR2 signalosome, was examined in the signaling cascade of the EBV oncogene LMP1. An IKK2/TPL2-dependent pathway mediating JNK activation by LMP1 was newly described and shown to be important for the survival of EBV-transformed B-cells. Furthermore, TNIK and its homologue MINK1 are required for posttranslational protein modifications of TRAF6 in the LMP1 signaling cascade. An important role of TNIK was also found for the process of migration of breast cancer cells, which has potential relevance for tumorigenesis.
