This thesis work focused on the phenotypic characterization of the ENU mouse mutant line TUB006. Heterozygous mutants present a selective T cell defect and susceptibility to Listeria infection. Homozygous mutants develop severe combined immunodeficiency (SCID), lipodystrophy, and systemic sterile autoinflammation. The underlying point mutation results in a single amino acid exchange in MECL-1, a catalytic subunit of the immuno- and thymoproteasome. The immunological phenotypes are mainly caused by selective cell death due to defective immuno- and thymoproteasome assembly.
