Die cGMP-abhängige Proteinkinase I (cGKI) ist ein Mediator der NO/cGMP-vermittelten Relaxation der vaskulären glatten Muskelzelle (VSMC). Die VSMC exprimiert zwei Isoformen der cGKI, cGKIalpha und cGKIbeta. In dieser Arbeit wurde mit Hilfe einer Gendeletions-/Rekonstitutions-Strategie die Rolle der cGKI-Isoformen bei der Regulation der zytosolischen Calcium-Konzentration über den NO/cGMP-Signalweg in VSMCs der Mausaorta untersucht. Es konnte gezeigt werden, dass in kultivierten cGKIalpha/beta-defizienten VSMCs ein Noradrenalin-induzierter Calcium-Transient in Anwesenheit des cGMP-Analogons 8-Br-cGMP nicht unterdrückt wird. Die defekte Calcium-Regulation in cGKI Knock-out VSMCs konnte durch Transfektion eines Fusionskonstruktes bestehend aus cGKIalpha und EGFP (enhanced green fluorescent protein) wiederhergestellt werden, aber nicht durch ein cGKIbeta-EGFP-Konstrukt. Dies weist auf cGKIalpha als eine Komponente bei der NO/cGMP-induzierten Relaxation der VSMC hin.     «

The cGMP-dependent protein kinase type I (cGKI) is a mediator of NO/cGMP-induced relaxation of vascular smooth muscle cells (VSMCs). VSMCs express two isoforms of cGKI, cGKIalpha and cGKIbeta. In this thesis a genetic deletion/reconstitution strategy was used to analyze the role of cGKI isoforms in the regulation of cytosolic calcium concentration by NO/cGMP in mouse aortic VSMCs. It was shown that in cultured VSMCs genetically deficient for cGKIalpha and cGKIbeta a noradrenaline-induced calcium transient is not suppressed in the presence of the cGMP analogue 8-Br-cGMP. The defective calcium regulation in cGKI knock-out VSMCs could be rescued by transfection of a fusion construct consisting of cGKIalpha and EGFP (enhanced green fluorescent protein) but not by cGKIbeta-EGFP. This suggests cGKIalpha to be a component of NO/cGMP-induced relaxation in VSMCs.     «
