The secretome is a major driver of inter-cellular communication. Mass spectrometry-based proteomics has the potential to decipher the secretome and identify quantitative alterations in disease conditions. However, until today proteomic secretome analysis remains technically challenging. In order to perform secretome analysis more efficiently, we developed the hiSPECS method. hiSPECS allows miniaturization of secretome analysis of primary cells in the presence of serum supplements and the determination of the substrate repertoire of proteases such as ADAM17 and BACE1.
