In eukaryotic cells, molecular motors provide means for directed long-range transport. Kinesin-2 is a heterodimeric motor that is specialized for intraflagellar transport in cilia. Using a fluorescence based super-resolution tracking technique, the special stepping behavior of these motors are examined. The results herein show the motors special adaption for the transport in cilia, previously unknown information on the asymmetric auto-inhibition mechanism and a difference in the individual stepping behaviors of the individual heads. This work significantly advances the understanding of heterodimeric kinesins and kinesins in general.
