The key component of the ubiquitin-mediated degradation pathway, the 20S proteasome, aids in the maintenance of cellular biological homeostasis and has received considerable attention for the treatment of many diseases, including cancer. In this work, the proteasome complexed with the natural product Vibralactone, and with the Belactosin A analogue were biochemically, functionally and structurally characterized. Additionally, the biochemical and structural analysis of the caseinolytic protease P (ClpP) was carried out, which is an important virulence regulator in multiresistant Stapylococcus aureus, making this structural data of high importance in the development of new inhibitors against pathogen bacteria.
