Beside their importance in medicine, antibodies are ideal model proteins for basic studies in structure formation and folding of secretory proteins. In this thesis, E. coli knock out strains were compared concerning their ability to produce active Fab fragments of a monoclonal antibody in the cytoplasm. Further, the in vitro folding of this Fab fragment and the influence of folding helpers on the folding process were investigated in detail. In this context, the cooperation between the ER resident Hsp70 homologue BiP and the protein disulfide isomerase (PDI) was analyzed. The results of this folding studies as well as the dissection of the ATPase cycle of BiP allow us deeper insights in the folding of secretory proteins in the ER and the function and regulation of molecular chaperones.
