Hypoplastic left heart syndrome (HLHS) is the most common and severe manifestation of a spectrum of congenital left ventricular outflow tract obstruction defects. Here, we performed induced pluripotent stem cells-based modeling, derived from HLHS patients, that demonstrated intrinsic defects in the cell-cycle/ciliogenesis/UPR-autophagy hub resulting in disrupted differentiation of early cardiac progenitor of first heart field and second heart field lineages. This evidence suggests that the primary onset of the disease occurs at the initial stages of cardiogenesis.
