PDAC presents an extensive desmoplastic reaction, composed mainly of CAFs. To study the role of CAFs in PDAC, a murine cohort was analyzed using multiplex IF, phenotyping CAFs populations, and characterizing stromal architecture. The stratification according to oncogenic-driver, differentiation, and Trp53 status uncovered context-specific activation patterns and different stromal organizations. The correlation between histopathological features and gene expression revealed an association between genetic background, ECM, CAFs activation, and increased metastasis frequency.
