External environmental factors are known to have an impact on immune functions. However, ionic tissue factors have thus far been underappreciated in the immune setting. We found that sodium chloride (NaCl) strengthens the Th2-phenotype in naïve and differentiated Th-cells and is notably concentrated in skin lesions of atopic dermatitis patients. Furthermore, NaCl is able to induce pro- and anti-inflammatory properties in Th17 cells, in tandem with the cytokine micromilieu. We identified IL1beta and TGFbeta as critical switch factors determining the dichotomous fate that NaCl exerts on T cells, thereby modulating the disease course of multiple sclerosis in mice.
