This thesis examined the possible mechanisms involved in leptin blood-brain-barrier transport and components of hypothalamic gliosis, which are both implicated in leptin resistance and obesity. Moreover, low dose childhood cranial irradiation, which is a potential novel risk factor for the metabolic syndrome, was investigated in a 2-year longitudinal murine study. Infrared labelled leptin tracing revealed no deficit in leptin transport, while careful analysis of gliosis reported astrocytosis after profound weight loss, but not in long term obesity. Furthermore, latent manifestations of the metabolic syndrome were revealed after moderate doses of cranial irradiation.
