Obesity and comorbid sequelae such as diabetes type 2 are global health burdens of the first order. The thesis aims to address three topics that focused on central vs. peripheral mechanisms of weight and glucose control. First, celastrol is identified as weight loss agent that requires functional hypothalamic leptin signaling. Second, the amelioration of HFD-induced hypothalamic astrocytosis is observed in calcineurin A beta deficient mice and third, hypothalamic GPx7 deletion is shown to induce diet-induced glucose intolerance.
