High HDAC2 expression in gastric cancer is associated with poor prognosis and is an independent prognostic marker. In this study, HDAC2 overexpression in human gastric cancer cell lines, tumor samples and in gastric tumors from CEA/Tag mice was shown. This overexpression provides a strong rationale for the use of HDACi as therapeutics. For the novel histone deacetylase inhibitor panobinostat (LBH589), molecular mechanisms underlying their anticancer effects could partially be determined. In the present study combination therapy by pretreatment with LBH589 followed by chemotherapeutic drugs proved a distinct superiority over the corresponding monotherapies in vitro and in vivo.
