SV40 LT-antigen is a viral oncoprotein with the ability to induce tumors by inhibition of tumor suppressors such as p53 and pRB. In this study we present evidence for a role of LT-antigen in suppressing the activity of CRL7 in mediating the ubiquitin-dependent degradation of insulin receptor substrate 1 (IRS1). This LT-mediated protection of IRS1 led to enhanced activation of promitogenic downstream signaling pathways AKT and Erk MAPK, which may contribute to viral-induced oncogenic transformation.
