Gamma-Butyrolactones were investigated for their protein reactivity and inhibition of bacterial virulence. This was achieved by synthesis of gamma-lactone probes for activity based protein profiling (ABPP), investigation of their protein reactivity using ABPP, the identification of bound proteins and the elucidation of the molecular mechanism of virulence inhibition. Two gamma-lactone probes showed a potent reduction of hemolysis and intracellular replication of Staphylococcus aureus by inhibition of the bacterial transcriptional regulators SarA, SarR and MgrA.
