Abstract:
The MAPT gene, encoding the microtubule-associated protein tau, is characterized by two allelic variants (H1 and H2) The more common haplotype H1/H1 is associated with an increased risk for several tauopathies, but also for the synucleinopathy Parkinson's disease (PD). The present study aimed to clarify whether the MAPT haplotype influences gene expression of tau and alpa-synuclein on mRNA and protein levels in postmortem brains of sporadic PD patients and controls. mRNA expression levels of several other MAPT haplotype-encoded genes were assessed.

Das MAPT-Gen, das für das Mikrotubuli assoziierte Protein Tau kodiert, ist durch zwei Allelvarianten (H1 und H2) charakterisiert. Der häufigere Haplotyp H1/H1 ist mit einem erhöhten Risiko für verschiedene Tauopathien, aber auch für die Synucleinopathie Morbus Parkinson (PD) verbunden. Diese Studie untersuchte, ob der MAPT-Haplotyp die Genexpression von Genen im MAPT Locus, Tau und Alpa-Synuclein auf mRNA- und Proteinebene in postmortem Gehirnen von sporadischen PD und Kontrollen beeinflusst.
