Abstract:
In this study, proteomics and transcriptomics studies of Thermoplasma acidophilum were performed. Mass spectrometry-based proteomics approaches combined with different pre-separation methods were used, to provide a global overview of the cytoplasmic proteins of T. acidophilum and information of proteins involved in the formation of macromolecular complexes. Additionally, a comprehensive quantitative proteomics and transcriptomics analysis of T. acidophilum cultured under aerobic and anaerobic conditions was applied which gave us information of biochemical adaptations of the cell to anaerobiosis. The results of this study are an important milestone for the validation of visual protein-maps of T. acidophilum created by cryo-electron tomography.

In dieser Arbeit wurden Proteom- und Transkriptom-Untersuchungen an Thermoplasma acidophilum durchgeführt. Für die Proteomanalyse wurden massenspektrometrische Verfahren in Kombination mit unterschiedlichen Vortrennungsmethoden benutzt, um einen globalen Überblick über die exprimierten cytosolischen Proteine von T. acidophilum und ein umfassendes Inventar der makromolekularen Komplexe zu erhalten. Zusätzlich wurde eine vergleichende Quantifizierung des Proteoms und des Transkriptoms von T. acidophilum, kultiviert unter aeroben und anaeroben Bedingungen, durchgeführt, welche Informationen über die biochemische Anpassung der Zelle an Anaerobiose lieferte. Die Ergebnisse dieser Studie sind ein wichtiger Meilenstein für die Validierung von mittels Kryo-Elektronentomographie erstellter visueller Proteinkarten von T. acidophilum.
