Protein kinase G (PknG) mediates survival of pathogenic mycobacteria in host macrophages. Thus it is a promising target for new tuberculosis therapies. Deletions or mutations in the N-terminal unstructured region (NORS) and the redox-sensitive rubredoxin-like domain (RD) significantly reduce the survival function. The aim of this study was the structural and dynamic characterization of these N-terminal regions by NMR spectroscopy and complementary methods. This provided new insights in how the NORS and the RD regulate the kinase activity.
