The beta secretase BACE1 is a central drug target in Alzheimer's disease therapy. The aim of this work was to analyze, if the transmembrane proteins PTPRN2, PLXDC2 and ENPP5 are substrates of the protease BACE1. For this reason, the proteins were recombinantly expressed in HEK293 cells and analyzed in a protease in-vitro-assay. These experiments showed that PTPRN2, PLXDC2 and ENPP5 are substrates of BACE1 in vivo and in vitro. Additionally, it could be shown that ENPP5 is a BACE1 substrate in primary murine neurons.
