In this study, the role of NOTCH1 and the tumor microenvironment of head and neck squamous cell carcinoma (HNSCC) was investigated. In vitro NOTCH1 inhibition impeded migration and proliferation of HNSCC and a high expression of NOTCH1 intracellular domain (NICD) in HNSCC patients was associated with reduced overall survival. The combination of tumor infiltrating lymphocytes (TILs) and NICD separated the patients into different clinical risk groups. Particularly in high-risk HNSCC with a low number of TILs, NOTCH1 could be a therapeutic target.
