The subject of this PhD thesis is the cloning of genes ispG and ispH of the deoxyxylulose phosphate pathway from the plants Solanum lycopersicum and Zea mays as well as the biochemical characterization of recombinant IspG proteins from Aquifex aeolicus, Bacillus subtilis, Haemophilus influenzae and Mycobacterium tuberculosis and of the recombinant IspH protein from Thermotoga maritima. The IspG protein from H. Influenzae and the IspH protein from T maritima were used to synthesise (E)-4-hydroxy-3-methylbut-2-enyl diphosphate and isopentenyl diphosphate/dimethylallyl diphosphate, respectively, from 2-C-methyl-D-erythritol-2,4-cyclodiphosphate in high amounts to use as substrate.
