In this pan-cancer study, 32 patients with 25 tumor types underwent comprehensive analysis, integrating proteogenomics for neoantigen discovery with phenotypic and functional analysis. Irrespective of entity, 90 neoantigens (32 validated) were identified, predominantly from RNA variants. 8 validated neoantigens showed in vitro immunogenicity. This study underscores the significance of RNA-derived variants in neoantigen identification and offers insights for enhancing future clinical applications.
