In this thesis, NMR structural and biochemical studies were conducted with the β-barrel membrane proteins OEP21, a chloroplast outer envelope protein, and hVDAC1, a mitochondrial outer membrane protein. Using NMR spectroscopy, the structure of OEP21 could be determined, enabling its functional characterization by biochemical assays and MD simulations. Furthermore, a novel method was established to quantify the membrane protein insertion into phospholipid nanodiscs using a GB1-fusion protein approach and hVDAC1 as a model system.
