Ziel dieser Arbeit war die Identifizierung und Charakterisierung von MHC-Klasse-I-Peptiden auf malignen hämatologischen Zellen im Hinblick auf ihre Eignung für Peptid-spezifische Immuntherapien. Hierzu wurden Leukämiezellen von acht Patienten mit Hilfe eines immunopeptidomischen Ansatzes untersucht. Durch Peptid-/MHC-Klasse-I Immunpräzipitation und anschließender massenspektrometrischer Analyse wurden potentielle Tumorantigene identifiziert und in weiteren Schritten validiert. Abschließend kann festgehalten werden, dass es sich bei einigen der identifizierten Tumorantigene um immunogene Zielstrukturen handelt, welche für passive Immunisierungsansätze, insbesondere im allogenen Kontext geeignet wären.     «

The aim of this study was the identification and characterization of class I HLA peptides presented on malignant hematopoietic cells in terms of their suitability for peptide-specific immunotherapies. Therefore, primary leukemia cells from eight patients were analyzed by an immunopeptidomic approach. After peptide/HLA class I immunoprecipitation and consecutive masspectrometric measurment, potential tumor antigens were identified and further validated. In conclusion, some of the identified tumor antigens might serve as targets for passive antitumor immunization approaches, especially in an allogeneic setting.     «
