The aldehyde dehydrogenase 1 (ALDH1A1) was identified as first and major protein target of duocarmycins via ABPP and LC/MS-MS. The role of ALDH1A1 in the cytotoxicity of duocarmycins as well as in lung cancer cell proliferation was clarified by inhibition and proliferation assays, cell imaging and siRNA knock down. In addition a protocol for the proteomic labeling of intracellular L. monocytogenes in murine macrophages was established and validated, which contributes to the identification of pathogenesis-associated enzymes that are not detected at an extracellular life cycle.
