The epidermal growth factor receptor 3 (HER3) is a tumor associated antigen, which is e.g. overexpressed in breast cancer and associated with a poor prognosis in these patients. In the context of this thesis, HER3356-364-specific T cell clones were generated as a source for a T cell receptor (TCR) gene transfer in primary T lymphocytes from breast cancer patients. For that purpose, CD8+ T cells from HLA-A2 negative donors were isolated after stimulation with HLA-A2+ HER3356-364-peptide loaded dendritic cells and cloned. The HER3356-364-specific T cell clones were able to recognize HLA-A2+ HER3+ breast cancer and melanoma cell lines in cytokine secretion and lysis assays.
