Implantatverluste sind hauptsächlich Folge mangelnder Osseointegration und bakterieller Infektionen. Ziel der Arbeit war es, osteoinduktiv und antiinfektiv beschichtete Titanimplantate in vitro auf ihre Wirksamkeit und Zellverträglichkeit zu prüfen. Es konnte ein nicht-viraler Gentransfer in 3T3-Fibroblasten und 293-Zellen mit Copolymer protected gene vectors (COPROGs) und PDLLA als Träger gezeigt werden, in das das Plasmid des osteoinduktiven Wachstumsfaktors BMP-2 integriert war. Mit Metridia-Luciferase als Reportergen in COPROGs wurde die Transfektion von humanen Osteoblasten, 3T3-Fibroblasten und mesenchymalen Stammzellen gezeigt. Die antiinfektiven Beschichtungen Chlorhexidindiglukonat und Triclosan hatten einen signifikant proliferationshemmenden Effekt auf humane Osteoblasten und 3T3-Fibroblasten sowie eine deutliche Veränderung der Zellmorphologie zur Folge. Derartige Beschichtungen könnten in vivo die Osseointegration fördern.
