Cellular c-Src and oncogenic v-Src kinase differ in their regulation and stability. It is unknown, which mutations are responsible fort these differences. In this thesis, it could be shown that only a combination of a triple mutation and the deletion of the C-terminal tail caused instability and an increase in activity of c-Src. Similar to v-Src, these mutations caused a lethal phenotype in yeast. Hence, only a combination of point mutations and the deletion are responsible for the differences in stability between c-Src and v-Src. Another difference is the influence of the molecular chaperone Hsp90 on Src. While c-Src is not affected, v-Src can be stabilized at low temperatures by Hsp90 only while further chaperones are necessary for stabilization at higher temperatures.
