Immuno Positron Emission Tomography (iPET) is a high sensitive imaging methodology based on the use of target-specific radiotracers made of proteins, such as antibodies and their derivatives, used to perform high selective in vivo disease diagnosis. Such tracers are based on a protein construct conjugated with either a prosthetic group or a chelator in order to carry the radioisotope of choice ideal to perform iPET. The aim of this thesis was to develop novel and more stable chelators to expand the possibilities of protein labellings in order to be used in iPET.

Die Immuno-Positronen-Emissions-Tomographie (iPET) ist ein hochempfindliches bildgebendes Verfahren, das auf der Verwendung von zielspezifischen Radiotracern aus Proteinen wie Antikörpern und deren Derivaten beruht und zur hochselektiven In-vivo-Diagnose von Krankheiten eingesetzt wird. Solche Tracer basieren auf einem Proteinkonstrukt, das entweder mit einer prosthetischen Gruppe oder einem Chelatbildner konjugiert ist, um das Radioisotop der Wahl zu tragen, das für die Durchführung der iPET ideal ist. Ziel dieser Arbeit war es, neuartige und stabilere Chelatoren zu entwickeln, um die Möglichkeiten der Proteinmarkierung für den Einsatz in der iPET zu erweitern.
