In this project affinity proteomics with different viral nucleic acids (NA) was used to identify conserved NA interacting proteins in different species (human, mouse and fly). 90 candidates were selected for functional characterization: measuring the replication 4 distinct in viruses identified 43 viral restriction factors and 13 proteins that supported virus growth. TAOK2, was selected for follow-up experiments, as its NA interaction was conserved and it showed a broad antiviral phenotype. We observed that the TAOK2 fly orthologue TAO kinase is a high affinity dsRNA binder and TAOK2 regulates IRF3-dependent antiviral responses without affecting NF-kB-dependent cytokine expression.

In diesem Projekt wurde Affinitätsproteomik mit viraler Nukleinsäuren (VN) verwendet, um konservierte VN-interagierende Proteine in drei Spezies (Mensch, Maus und Fliege) zu entdecken. 90 Kandidaten wurden für die funktionelle Charakterisierung ausgewählt: Durch Replikationsmessung von 4 Viren wurden 43 virale Restriktionsfaktoren und 13 provirale Proteine identifiziert. TAOK2 wurde für Folgeexperimente ausgewählt, da seine VN-Interaktion konserviert ist und es einen breiten antiviralen Phänotyp hat. Wir beobachteten, dass TAOK2-Fliegenortholog TAO Kinase ein hochaffiner dsRNA Interaktor ist und TAOK2 IRF3-abhängige antivirale Reaktionen reguliert, ohne NF-kB-abhängige Cytokinexpression zu beeinflussen.
