Abstract:
Eine neue differenzielle off-line Kopplung von HPLC mit NMR-Spektroskopie (DOLC-NMR) wurde entwickelt, um eine vergleichende Metabolomanalyse durchzuführen. Die Veränderungen im Metabolom von S. cerevisiae und P. roqueforti wurden vor und nach der Intervention mit aromatischen Aminosäuren erfasst. Im Metabolom der Hefe konnten erstmals N-(1-Oxoacyl)-L-tyrosin Derivate detektiert werden, welche bei humansensorischen Experimenten eine Modulation des kokumi Effekts hervorriefen. Die Aufklärung der Biosynthese dieser und weitere Hefemetaboliten gelang mittels 13C Markierungsexperimenten. Im Metabolom von Penicillium roqueforti konnten erstmals drei neue Metabolite identifiziert werden, welche antimikrobielle Eigenschaften zeigten. Der Gehalt dieser und weiterer Sekundärmetaboliten aus P. roqueforti wurde im Rahmen einer Warenkorbanalyse von Blau- und Weißschimmelkäse untersucht.

A novel Differential Off-line LC-NMR approach (DOLC-NMR) was developed to capture and quantify nutrient-induced metabolome alterations in S. cerevisiae and P. roqueforti upon interventions with aromatic amino acids. Among the metabolites identified in the metabolome of S. cerevisiae, N-(1-oxoacyl)-L-tyrosine derivatives imparted a kokumi taste enhancement in sensory experiments. Labelling experiments using 13C confirmed the biosynthetic pathway leading to key metabolites. The metabolome analyses of P. roqueforti revealed three metabolites reported for the first time as fungi metabolites. Antimicrobial activity tests of identified substances showed strong inhibitory effects on yeast or bacteria cells. In addition, selected secondary metabolites were quantified in commercially available white and blue-mold cheese samples.
