We isolated Hodgkin and Reed-Sternberg cells by single cell microdissection, performed microarray analyses and compared their expression signatures to those of germinal center B cells. MLL5 was identified as overexpressed in Hodgkin lymphoma (HL), and knockdown of MLL5 in HL cell lines resulted in a significantly decreased proliferation, an increased apoptosis after growth factor withdrawal and an enhanced responsiveness towards chemotherapy agent etoposide. Moreover, Western blot analyses revealed a potential role of MLL5 in the epigenetic regulation of gene activation in HL cells.
