In this work protein multifunctionality was investigated in three representatives of the enzyme family of 17beta-hydroxysteroid dehydrogenases (17beta-HSDs). A new retinoid reducing capability was discovered in 17beta-HSD1, which was up to now only known to catalyze the coversion of estrogens. Additionally, an amino acid crucial for the discrimination of steroid and retinoid substrates was identified. Studies on the role of the enzyme 17beta-HSD12 in steroid and fatty acid metabolism were conducted. Furthermore, a new multifunctionality was discovered in rodent 17beta-HSD3. The development of a LC-MS/MS method for steroid analysis allowed insights into the metabolism of these hormones.
