- Abstract:
- Ziel der Arbeit war zunächst die Entwicklung eines ELISA zum quantitativen Nachweis humaner HER2/neu-Antikörper und dann die Untersuchung von Seren von Patienten mit Mammatumoren auf mögliche autologe HER2/neu -Antikörper. Mit dem entwickelten ELISA konnten keine autologen IgG-/IgM-Antikörper gegen HER2/neu in humanem Seren (n=173) nachgewiesen werden. Aber es gelang zuverlässig, das Immunotherapeutikum Herceptin® im Serum und anderen Körperflüssigkeiten quantitativ nachzuweisen.
- Translated abstract:
- The objective of this thesis was the development of an Enzyme Linked Immunosorbent Assay (ELISA) to quantify human HER2/neu antibodies and afterwards the screening of sera from patients with breast tumours for possible autoantibodies to HER2/neu. It was not possible to detect autoantibodies (IgG/IgM) in human sera (n= 173) with this ELISA, but the immunotherapy Herceptin® was reliably quantified in serum and other body fluids, an important basis for the ELISA’s use in clinical research.
