Abstract:
Drei Proteinkomponenten des Maltose/Maltodextrin-Systems von E. coli wurden biochemisch und kristallographisch charakterisiert. 72 verschiedene Konstrukte des Regulators MalT wurden hinsichtlich Exprimierbarkeit, Stabilität und Oligomerzustand durchmustert. Zum Verständnis des molekularen Mechanismus der durch MalQ katalysierten Glycosyltranferase-Reaktion wurden Kristallstrukturen der apo-Form sowie Komplexe mit Maltose bzw. einem Inhibitor mit Auflösungen von bis zu 2,1 Å aufgeklärt. Von MalM wurden die Signalpeptidsequenz ermittelt und die N- und C-terminale Domänen kristallisiert.

Three protein components of the maltose/maltodextrin system in E. coli were biochemically and crystallographically characterized. 72 different constructs of the regulator MalT have been screened with regard to expression yield, stability and oligomerization tendency. To understand the molecular mechanism of the glycosyltranferase reaction catalyzed by MalQ crystal structures of the apo form and complexes with maltose and an inhibitor were elucidated at resolutions down to 2.1 Å. The signal peptide of MalM was determined and crystals of its N- or C-terminal domains were obtained.
