Abstract:
Die Präimplantationsdiagnostik (PID) ermöglicht Paaren, in deren Familien eine schwere monogene Erkrankung aufgetreten ist, das Risiko für die Geburt eines Kindes, das von dieser Erkrankung betroffen ist, nahezu vollständig auszuschließen. Der Schwerpunkt dieser Dissertation war es, ein familienspezifisches Testsystem für Trophektoderm-Biopsate bei PID auf der Basis der Qualitätsrichtlinien der Europäischen Gesellschaft für humane Reproduktion und Embryologie (ESHRE) für eine Familie, die von der monogenen Erkrankung Osteogenesis imperfecta Typ I betroffen ist, zu erstellen und zu validieren. Die Änderung des Embryonenschutzgesetzes im Jahr 2011 durch Einführung des §3a ermöglichte die PID in Deutschland.

Preimplantation genetic diagnosis (PGD) enables couples, whose families are affected by a monogenic disease, to almost exclude the probability of birth of an affected child. The objective of this thesis was to establish and validate a PGD assay compatible with the ESHRE guidelines (European Society for Human Reproduction and Embryology) for a specific couple affected by a severe phenotype of the monogenic disease Osteogenesis imperfecta type I. In 2011 paragraph §3a was implemented in the embryonic protection law that enabled PGD in Germany.
