Abstract:
HGPS is a premature aging disease, caused by a mutation in LMNA. The mutated Lamin A is called progerin and causes several cellular defects e.g., nuclear dysmorphism and Nuclear Pore Complex clustering (NPCs). To identify why NPCs cluster in HGPS, I evaluated both postmitotic NPC reassembly and NPC clustering in senescent control/HGPS cells. I could not find any defects in mitosis and observed clustering only in dysmorphic control/HGPS nuclei. I determined that NPC distribution needs an intact nuclear envelope and any changes, due to progerin or senescence, can result in clusters.

HGPS ist eine Krankheit des vorzeitigen Alterns, die durch eine Mutation in LMNA verursacht wird. Das mutierte Lamin A heißt Progerin und verursacht mehrere zelluläre Defekte, z. B. Kerndysmorphismus und Anhäufung von Kernporenkomplexen (NPCs). Um herauszufinden, warum sich NPCs in HGPS anhäufen, habe ich die postmitotische NPC-Neubildung und Anhäufung in seneszenten Kontroll-/HGPS-Zellen untersucht. Ich fand keine mitotischen Defekte und beobachtete die Anhäufung nur in dysmorphen Kontroll-/HGPS- Zellkernen. Ich stellte fest, dass die Verteilung eine intakte Zellkernmembran braucht und dass Veränderungen, durch z. B. Progerin oder Seneszenz, zu Clustern führen können.
