Frauen mit BRCA1/2 Mutation sind Hochrisikopatientinnen für Brust- u/o Eierstockkrebs, für die es neue Präventionsstrategien zu etablieren gilt. Der OPG/RANK/RANKL Signalweg spielt eine entscheidende Rolle in der Mammakarzinogenes, ist bei gBRCA1/2 Mutation hyperaktiviert und lässt sich durch Gesundheitsverhalten modulieren. Es konnte gezeigt werden, dass die Einhaltung einer mediterranen Ernährung und regelmäßige körperliche Aktivität im Rahmen der LIBRE-1 Lebensstilintervention zu einer signifikanten Erhöhung von OPG und Reduktion von sRANKL führt.

Women with a gBRCA1/2 mutation are high-risk patients for breast and/or ovarian cancer, for which new prevention strategies are needed. The OPG/RANK/RANKL pathway plays an important role in mammary carcinogenesis, is hyperactivated in BRCA1/2 mutation carriers and has been found to be affected by a healthy lifestyle. The present dissertation demonstrated that physical activity and adherence to the Mediterranean diet within the LIBRE-1 lifestyle intervention can significantly increase OPG and reduce sRANKL.
