In this thesis, two non-invasive methods have been developed for longitudinal expression analysis of alternatively spliced proteins and non-coding RNAs. We developed an exon-specific isoform expression reporter system (EXSISERS) that reports a specific exon-containing isoform of an endogenous gene by scarlessly excising reporter proteins post-translationally from the host protein. Also, we developed an intron-specific exon-independent coding transcript system (INSPECT) based on a reporter modality hidden in an engineered intron, allowing scarless transcriptional coupling of the reporter to the non-coding host gene.

In dieser Arbeit wurden zwei nicht-invasive Methoden zur longitudinalen Expressionsanalyse von alternativ gespleißten Proteinen und nicht-kodierenden RNAs entwickelt. Wir haben ein exonspezifisches Isoform-Expressions-Reportersystem (EXSISERS) entwickelt, das eine spezifische exonhaltige Isoform eines endogenen Gens anzeigt, indem es Reporterproteine posttranslational aus dem Wirtsprotein narbenlos herausschneidet.
