Abstract:
Die Beta-Sekretase BACE1 stellt einen zentralen Ansatzpunkt in der Theapie der Alzheimer-Krankheit dar. Das Ziel dieser Arbeit war zu untersuchen, ob die Transmembranproteine PTPRN2, PLXDC2 und ENPP5 Substrate von BACE1 sind. Hierfür wurden die Proteine rekombinant in HEK293-Zellen exprimiert und in einem Protease-in-vitro-Assay untersucht. Durch diese Experimente konnten PTPRN2, PLXDC2 und ENPP5 als Substrate der Alzheimer-Protease BACE1 validiert werden. Außerdem konnte ENPP5 in primären murinen Neuronen als BACE1-Substrat validiert werden.

The beta secretase BACE1 is a central drug target in Alzheimer's disease therapy. The aim of this work was to analyze, if the transmembrane proteins PTPRN2, PLXDC2 and ENPP5 are substrates of the protease BACE1. For this reason, the proteins were recombinantly expressed in HEK293 cells and analyzed in a protease in-vitro-assay. These experiments showed that PTPRN2, PLXDC2 and ENPP5 are substrates of BACE1 in vivo and in vitro. Additionally, it could be shown that ENPP5 is a BACE1 substrate in primary murine neurons.
