Abstract:
The present doctoral thesis investigates RIPK3-dependent inflammatory cell death (necroptosis) as a tumor suppressor in lung adenocarcinoma (LUAD). We evaluated RIPK3-associated inflammation and immune cell signatures on gene expression levels using mRNA-sequenced LUAD datasets from “The Cancer Genome Atlas” project. We delineated the phenotypical impact of RIPK3 expression on tumor outgrowth in genetically engineered mouse models for LUAD and evaluated different chemotherapeutics applied in LUAD regimens for inducing RIPK3-dependent necroptosis offering a potential treatment target.

Vorliegende Doktorarbeit untersucht den RIPK3-abhängigen inflammatorischen Zelltod im Adenokarzinom der Lunge (LUAD) mit dem Ziel der Identifikation assoziierter tumorsuppressiver und immunmodulatorischer Effekte. Dabei wurden mRNA-Sequenzierungen der LUAD-Datensätze des „The Cancer Genome Atlas“ Projekts analysiert und die RIPK3 Expression mit inflammatorischen Genprofilen und Immunzellsignalen korreliert. Zudem wurden genetisch modifizierte Mausmodelle etabliert, um das Tumorwachstum in Abhängigkeit von der RIPK3 Expression zu untersuchen. Ferner wurden Chemotherapeutika nach ihrer Fähigkeit, Nekroptose zu initiieren, evaluiert, um einen möglichen Therapieansatz aufzudecken.
